目的探讨重组胸腺素α1(Recombinant human thymosinα1,rhTα1)对免疫低下小鼠接种流感疫苗后免疫应答效果的影响。方法将BALB/c小鼠随机分为5组:疫苗组、环磷酰胺+疫苗组、环磷酰胺+疫苗+rhTα1低浓度(0.2 mg/kg)组、环磷酰胺+疫苗+rhTα1中浓度(0.4 mg/kg)组、环磷酰胺+疫苗+rhTα1高浓度(0.8 mg/kg)组。经小鼠腹腔注射环磷酰胺,80 mg/kg,共3次,隔天给药;肌肉接种流感疫苗,1.8μg血凝素/只;皮下注射低、中、高浓度rhTα1,0.2 ml/只,每周2次,连续4周。动态监测各组小鼠血清血凝抑制(HI)抗体效价,并观察小鼠脾脏指数及脾脏形态学结构的变化。结果与疫苗组相比,环磷酰胺+疫苗组小鼠体内HI抗体效价降低,脾指数降低,脾脏组织病理形态退化;与环磷酰胺+疫苗组相比,环磷酰胺+疫苗+rh-Tα1各浓度组HI抗体效价升高,小鼠脾指数增加,脾脏组织病理形态明显改善。结论重组胸腺素α1可提高免疫低下小鼠流感疫苗接种的免疫效果,为寻找适合免疫低下人群的疫苗增强剂提供了有益的尝试。
Objective To investigate the effect of recombinant human thymosin α1 (rhTα1) on the immune response in mice immunosuppressed with influenza vaccine. Methods BALB / c mice were randomly divided into 5 groups: vaccine group, cyclophosphamide + vaccine group, cyclophosphamide + vaccine + low concentration of rhTα1 (0.2 mg / kg), cyclophosphamide + 0.4 mg / kg), cyclophosphamide + vaccine + high concentration of rhTα1 (0.8 mg / kg). The mice were intraperitoneally injected with cyclophosphamide, 80 mg / kg, three times a day for administration; the flu was administered intramuscularly and 1.8 μg of hemagglutinin per mouse; hypodermic injection of low, medium and high concentrations of rhTα1 at 0.2 ml / mouse , 2 times a week, for 4 weeks. The serum hemagglutination inhibition (HI) antibody titers of mice in each group were dynamically monitored, and the changes of spleen index and morphology of spleen were observed. Results Compared with the vaccine group, HI antibody titer and spleen index of mice in the cyclophosphamide + vaccine group decreased and the histopathology of the spleen decreased. Compared with the cyclophosphamide + vaccine group, cyclophosphamide + vaccine + rh- The HI antibody titers in each concentration of Tα1 increased, the spleen index of mice increased, the histopathology of spleen significantly improved. Conclusion Recombinant thymosin α1 can enhance the immune effect of influenza vaccination in immunocompromised mice, and provide a useful attempt to find a vaccine enhancer suitable for immunocompromised people.
