目的:观察高原红细胞增多症患者骨髓红系祖细胞体外增殖情况。方法:采用微量甲基纤维素培养法对10例高原红细胞增多症(高红组)及10例高原健康人(对照组)的骨髓细胞进行体外培养。结果:在无促红细胞生成素刺激时,高红组及对照组皆无CFU-E及BFU-E集落形成。高红组的CFU-E及BFU-E集落数随促红细胞生成素的浓度增加而增加(P<0.01),当促红细胞生成素浓度达到8u/ml时,CFU-E及BFU-E的集落数不再明显增加(P>0.05),对照组对促红细胞生成素的刺激时反应与高红组相一致。在同一浓度促红细胞生成素刺激时,两组CFU-E及BFU-E集落数无明显差别(P>0.05)。结论:高原红细胞增多症患者骨髓红系祖细胞自身无增殖能力,其增殖必须有促红细胞生成素的刺激,促红细胞生成素的增高是高原红细胞增多症发病的一个因素,但其发病还存在其他机制。
Objective: To observe the proliferation of bone marrow erythroid progenitor cells in patients with high altitude polycythemia. Methods: Bone marrow cells from 10 cases of high altitude polycythemia (high red group) and 10 cases of high altitude healthy people (control group) were cultured in vitro by using the method of micro-methylcellulose culture. Results: In the absence of erythropoietin stimulation, there were no CFU-E and BFU-E colony formation in the high-red group and the control group. The number of CFU-E and BFU-E colonies in high-red group increased with the increase of erythropoietin (P <0.01). When the erythropoietin concentration reached 8u / ml, CFU-E and BFU-E (P> 0.05). The response to erythropoietin in control group was consistent with that in Gaohong group. There were no significant differences in the number of CFU-E and BFU-E colonies between the two groups when stimulated with erythropoietin (P> 0.05). CONCLUSION: Bone marrow erythroid progenitor cells in patients with high altitude polycythemia have no proliferative ability. Erythropoietin must be stimulated by the proliferation of erythropoietin. Elevated erythropoietin is a factor in the pathogenesis of high altitude polycythemia, but there are other mechanism.
