目的 克隆制备人源性抗角蛋白 Ig M抗体 .方法 以人表皮角蛋白为抗原 ,从人源性天然抗体库中筛选 Ig M型抗角蛋白人抗体 ,并用 EL ISA对所获抗体进行抗原结合活性和特异性等鉴定 .结果 通过 3轮“亲和吸附 -洗脱 -扩增”筛选 ,获得 2株 Ig M型抗角蛋白噬菌体抗体 .酶切去掉载体上的噬菌体基因 ,得到了可溶性表达的 Fab段 .经过鉴定 ,所获抗体具有良好的抗原结合活性和特异性 .结论 从人源性天然抗体库中成功筛选出 2株抗角蛋白 Ig M抗体 ,为进一步研究抗角蛋白抗体的特性提供了条件
Objective To clone and prepare human anti-keratin Ig M antibody.Methods Human anti-keratin Ig M antibody was screened from human natural antibody library using human epidermal keratin antigen and the obtained antibody was antigen-conjugated Activity and specificity.RESULTS Two strains of IgM-type anti-keratin antibodies were screened by 3 rounds of “affinity-adsorption-eluting-amplification”, and the bacteriophage gene was cloned by enzyme digestion to obtain the soluble Fab fragment.After identification, the obtained antibody has good antigen binding activity and specificity.Conclusion Two anti-keratin Ig M antibodies were successfully screened from the human natural antibody library, which provide further evidence for the anti-keratin antibody properties Conditions
