目的:研究口腔鳞状细胞癌组织中Pokemon、Ku70的表达及其临床病理学意义。方法:采用免疫组化En-vision二步法检测Pokemon、Ku70在47例口腔鳞状细胞癌组、20例正常口腔黏膜组中的表达。结果:1)Pokemon在口腔鳞状细胞癌组及正常口腔黏膜组中阳性表达率分别为:68.1%(32/47)、25.0%(5/20),P<0.05。高分化组与中低分化组Pokemon的阳性表达率分别为45.0%、85.2%(P<0.05)。不同TNM的口腔鳞状细胞癌组织中,Ⅰ期+Ⅱ期组、Ⅲ期+Ⅳ期组Pokemon的阳性表达率分别为88.0%、45.5%(P<0.05)。有淋巴结转移组、无淋巴结转移组Pokemon的阳性表达率分别为86.7%、35.5%(P<0.05)。2)Ku70在口腔鳞状细胞癌组及正常口腔黏膜组中阳性表达率分别为:61.7%(29/47)、30.0%(6/20),P<0.05。高分化组与中低分化组Ku70的阳性表达率分别为55.0%、66.7%(P>0.05)。不同TNM分期的口腔鳞状细胞癌组织中,Ⅰ期+Ⅱ期组、Ⅲ期+Ⅳ期组Ku70的阳性表达率分别为72.0%、50.0%(P>0.05)。有淋巴结转移组、无淋巴结转移组Ku70的阳性表达率分别为73.3%、35.5%(P<0.05)。结论:提示Pokemon及Ku70在口腔鳞状细胞癌发生发展中起重要作用;Pokemon与Ku70可能成为判定口腔鳞状细胞癌预后的生物标志物。
Objective: To investigate the expression of Pokemon and Ku70 in oral squamous cell carcinoma and its clinicopathological significance. Methods: Immunohistochemical En-vision two-step method was used to detect the expression of Pokemon and Ku70 in 47 cases of oral squamous cell carcinoma and 20 cases of normal oral mucosa. Results: 1) The positive rates of Pokemon in oral squamous cell carcinoma and normal oral mucosa were 68.1% (32/47) and 25.0% (5/20) respectively (P <0.05). The positive rates of Pokemon in well-differentiated and moderate-poorly differentiated groups were 45.0% and 85.2%, respectively (P <0.05). The positive rates of Pokemon in stage Ⅰ + group and stage Ⅲ + Ⅳ group were 88.0% and 45.5% (P0.05) respectively in different TNM oral squamous cell carcinoma tissues. The positive rates of Pokemon in patients with lymph node metastasis and no lymph node metastasis were 86.7% and 35.5%, respectively (P <0.05). 2) The positive rates of Ku70 expression in oral squamous cell carcinoma and normal oral mucosa were 61.7% (29/47) and 30.0% (6/20) respectively, P <0.05. The positive expression rates of Ku70 in well-differentiated and moderate-poorly differentiated groups were 55.0% and 66.7%, respectively (P> 0.05). The expression rates of Ku70 in stage Ⅰ + stage Ⅱ and stage Ⅳ were 72.0% and 50.0% (P0.05), respectively, in different stages of oral squamous cell carcinoma. The positive expression rate of Ku70 with lymph node metastasis and without lymph node metastasis was 73.3% and 35.5% respectively (P <0.05). Conclusion: Pokemon and Ku70 play an important role in the development of oral squamous cell carcinoma. Pokemon and Ku70 may be biomarkers for prognosis of oral squamous cell carcinoma.
