运用各种柱色谱方法和制备型HPLC技术对柔毛鸦胆子茎的化学成分进行分离纯化,共从其茎的95%乙醇提取物中分离得到10个化合物,采用NMR等波谱技术鉴定其结构分别为deacetylated isobrucein B(1),indaquassin X(2),cleomiscosin A(3),cleomiscosin B(4),(+)-lyoniresinol(5),(+)-表松脂醇(6),(+)-松脂醇(7),(+)-丁香脂素(8),4,5-dihydroblumenol A(9),腺苷(10)。采用MTT法对以上化合物进行了体外细胞毒活性测试,化合物2对人源肿瘤细胞HT-29,HepG2,BGC-823和SKOV3具有显著的细胞毒活性(IC500.84~3.97μmol.L-1)。所有化合物均为首次从该植物中分离得到,其中化合物1是1个新天然产物。化合物2对人源肿瘤细胞HT-29,HepG2,BGC-823和SK-OV3具有显著的细胞毒活性。
A total of 10 compounds were isolated from the 95% ethanol extract of the stems by using various column chromatographic methods and preparative HPLC techniques. The structures of the compounds were identified by NMR and other spectroscopic techniques (+) - lyoniresinol (5), (+) - epihalohydrin (6), (+) - Pinoresinol (7), (+) - Syringalin (8), 4,5-dihydroblumenol A (9), Adenosine (10). The above compounds were tested for cytotoxicity by MTT method. Compound 2 showed significant cytotoxic activity on human tumor cells HT-29, HepG2, BGC-823 and SKOV3 (IC500.84 ~ 3.97μmol.L-1) . All compounds were isolated from this plant for the first time, of which compound 1 is a new natural product. Compound 2 has significant cytotoxic activity on human tumor cells HT-29, HepG2, BGC-823 and SK-OV3.
