目的 研究人卵巢癌组织中微小RNA-451(miR-451)的表达及对卵巢癌细胞增殖和凋亡的影响.方法 分别取对数生长期的miR-451及miR-NC细胞,制成细胞悬液,调整终浓度为2.0×107cell·mL-1.将0.2 cell·mL-1细胞悬液接种于裸鼠背部近右下肢处皮下组织内,建立裸鼠皮下成瘤模型.按照体重将5周龄BLAB/c裸鼠随机分为2组,每组3只:miR-451组及对照组.miR-451组加入miR-451慢病毒上清液1 mL、完全培养基2 mL和聚凝胺15μg;对照组加入miR-NC慢病毒上清液1 mL、完全培养基2 mL和聚凝胺15μg.5周后处死动物获取皮下肿瘤.以PCR法检测卵巢癌组织中miR-451表达水平;以流式细胞仪测定SKVO3细胞的凋亡比例;以裸鼠皮下种植瘤模型的构建测定肿瘤体积.通过qPCR和蛋白免疫印迹检测过表达miR-451后SKVO3细胞内ATF2表达的变化.结果 miR-451在卵巢癌组织和癌旁组织中的表达水平分别为0.39±0.03,1.25±0.03,前者明显低于后者,差异有统计学意义(P<0.05).过表达miR-451表达后,卵巢癌SKVO3细胞凋亡比例为(57.10±8.92)%,较对照组的(21.08±5.04)%明显升高,差异有统计学意义(P<0.01).miR-451组与miR-NC组的肿瘤体积分别为(0.40±0.08),(1.35±0.23) cm3,组间差异有统计学意义(P<0.01).在过表达miR-451的肿瘤组织中,ATF2的免疫组化染色强度显著降低,差异有统计学意义(P<0.05).结论 miR-451在卵巢细胞癌中表达降低,上调miR-451可抑制ATF2表达,以此发挥抗肿瘤生长作用.“,”Objective To explore the expression of microRNA-451 (miR-451) in human ovarian cancer tissue and its effect on ovarian cancer cell proliferation and apoptosis.Methods The miR-451 and miR-NC cells of logarithmic growth were taken respectively,and the cell suspension was made by cold PBS solution,and the final concentration was 2.0 x 107 cell · mL-1.The 0.2 cell · mL-1 were inoculated into the subcutaneous tissue of the lower right lower limb of the nude mouse,and the model of subcutaneous tumor was established.Six 5-week old BLAB/c nude mice were randomly divided into two groups:the miR-451 group and the control group,and each group had three nude mouse.In the mir-451 group,miR-451 slow virus supernatant 1 mL,complete culture medium 2 mL and ploybrene 15 g were added to each hole.In the control group,miR-NC slow virus supematant 1 mL,complete culture medium 2 mL and ploybrene 15 g were added to each pore.After 5 weeks of obtaining animal subcutaneous tumor,the expression level of miR-451 in ovarian cancer tissues was detected by Polymerase Chain Reaction.The apoptosis rate of SKVO3 cells was detected by flow cytometer.The gross tumor volume of nude mouse was detected through construct the nude mice subcutaneous tumor model.Changes of ATF2 expression in SKVO3 cells After overexpression of miR-451 was detected by RqPCR and Westen bloting.Results The expression level of miR-451 with ovarian cancer (0.39 ± 0.03) was significantly lower than that in the adjacent tissues of low cancer (1.25 ± 0.03),and the difference was statistically significant (P < 0.05).After expressing miR-451 expression,the apoptosis rate of SKVO3 cells in ovarian cancer (57.10 ± 8.92) %.Compared with the control group (21.08 ± 5.04),the difference was statistically significant (P < 0.01).The total tumor volume of miR-451 group (1.35 ± 0.23) cm3 was significantly lower than that in the mir-nc group (0.40 ±0.08) cm3,the difference was statistically significant (P <0.01).In the tumor tissue of overexpression level of miR-451,immunohistochemical staining intensity of activating transcription factor 2 (ATF2) was decreased significantly(P < 0.05).Conclusion The expression of miR-451 in ovarian cell carcinoma was decreased,and the upregulation of miR-451 could inhibit the expression of ATF2,thus playing a role in anti-tumor growth.
