本文应用免疫组化及PCR-SSCP技术对58例乳腺肿瘤c-erbB-2癌基因表达及p53抑癌基因突变进行研究,结果显示乳腺癌中c-erbB-2表达率为38%,p53表达率为52%,导管内乳头状瘤均为阴性;c-erbB-2表达与乳腺癌组织学分级(P<0.01)及淋巴结转移呈负相关(P<0.01),与雌激素受体状态呈正相关(P<0.01);p53表达与淋巴结转移呈正相关(P<0.01),且倾相于≥2cm的肿瘤;两基因的表达均与乳腺上皮细胞的恶性转化及淋巴结转移密切相关,但两基因间无明显相关性;因此,每一基因的改变均可作为不依赖于淋巴结状态的独立预后指标。PCR-SSCP分析表明,15例p53基因表达阳性病例中4例于外显子5,6例于外显子7有该基因的突变(67%),说明乳腺癌p53基因突变也多发生于其突变热点部位。
In this paper, 58 cases of breast cancer c-erbB-2 oncogene and p53 tumor suppressor gene mutations were studied by immunohistochemistry and PCR-SSCP technology. The results showed that the expression of c-erbB-2 was 38% in breast cancer and the expression of p53 The positive rate of c-erbB-2 was negatively correlated with histological grade (P <0.01) and lymph node metastasis (P <0.01), but not with intraductal papilloma (P <0.01). There was a positive correlation between the expression of p53 and lymph node metastasis (P <0.01), and the tumor size of ≥2 cm. The expressions of both genes were correlated with the malignant transformation of mammary epithelial cells and Lymph node metastasis is closely related, but no significant correlation between the two genes; therefore, each gene change can be independent of lymph node status independent prognostic indicators. PCR-SSCP analysis showed that in 15 cases of p53 gene expression positive cases, 4 cases of exon 5 and 6 cases of exon 7 mutations (67%), indicating that mutations in breast cancer p53 gene also occurred in Mutation hot spots.
