建立了一种快速简便直接检测双孢蘑菇培养料中微量1-氨基环丙烷-1-羧酸(ACC)含量的离子色谱法。此法使用DionexICS-3000离子色谱仪与AminoPacPA-10离子交换柱,以0.25mol·L-1NaOH、1.00mol·L-1NaAc和水作为洗脱液进行梯度洗脱,流速0.25mL·min-1,柱温30℃,进样量25μL。在此条件下,信噪比为3时,ACC的保留时间为9.284min,最低检测限是0.0330μg·mL-1,标准曲线线性符合系数为0.9990,线性范围在0.10 ̄2.00μg·mL-1。三个加标水平下的回收率分别为106.00%、91.71%和96.08%。该方法可用于检测双孢蘑菇培养料中的ACC含量,检测结果为3.6620μg·g-1(FW)。此法也适用于其他食用菌和土壤微生物的研究。
A rapid and simple method for direct determination of trace amounts of 1-aminocyclopropane-1-carboxylic acid (ACC) in Agaricus bisporus culture medium was established. The method uses DionexICS-3000 ion chromatograph and AminoPacPA-10 ion exchange column with gradient elution with 0.25mol·L-1NaOH, 1.00mol·L-1NaAc and water as eluent, the flow rate is 0.25mL · min-1, Column temperature 30 ℃, injection volume 25μL. Under this condition, the retention time of ACC was 9.284min with the signal to noise ratio of 3, the lowest detection limit was 0.0330μg · mL-1, the linearity coefficient of standard curve was 0.9990, the linear range was 0.10-2.00μg · mL-1 . The recoveries at the three spike levels were 106.00%, 91.71% and 96.08% respectively. The method can be used to detect the content of ACC in Agaricus bisporus culture material, and the test result is 3.6620 μg · g-1 (FW). This method is also applicable to other edible fungi and soil microorganisms.
