目的:探讨P19细胞向心肌细胞分化过程中微小RNA(miRNA)差异表达谱。方法采用miRNA芯片技术获得P19细胞分化前和分化后第8天的miRNA差异表达谱,定量PCR技术进行芯片结果验证,并采用生物信息学方法进行靶基因预测。结果共获得31条miRNAs差异表达谱。与未分化的P19细胞相比,分化第8天的细胞中16条miRNAs下调,15条上调。其中,miRNA‐32、miRNA‐25、miRNA‐21、miRNA‐363和miRNA‐367经定量PCR验证结果与芯片结果中的变化趋势一致。靶基因预测结果显示,GATA6、Notch1和 Wnt5a为miRNA‐32可能的靶基因。结论本研究获得了P19细胞向心肌细胞分化过程中miRNA差异表达谱,为先天性心脏病的研究提供了线索。“,”Objective To investigate the changes of microRNA (miRNA ) expression profiles during differentiation of P19 cells into cardiac myocytes .Methods The miRNA expression profile of P19 cells before and on the 8th day after differentiation was obtained by microarray .The quantitative PCR was used to verify the microarray results and bioinformatic method was used to predict the target genes .Results The miRNA microarray showed 31 differential expression miRNAs .Compared to before ,15 miRNAs were up‐regulated and 16 miRNAs were down‐regulated on the 8th day after differentiation .The quantitative PCR results of miRNA‐32 ,miRNA‐25 ,miRNA‐21 ,miRNA‐363 and miRNA‐367 were consistent with chip results .Bioinformatics method showed that GATA6 ,Notch1 and Wnt5a were the possible target genes of miRNA‐32 .Conclusion The changes of miRNA expression profiles during differentiation of P19 cells into cardiac myocytes have been obtained ,which could provide a clue to the study of congenital heart disease .
