目的探讨漆姑草水提物(HSJ)对人白血病细胞株K562细胞凋亡及细胞周期影响。方法采用四甲基偶氮唑蓝(MTT)比色法检测HSJ对K562细胞的增殖抑制作用;台盼蓝拒染法观察HSJ对细胞生长曲线影响;流式细胞仪检测HSJ对K562细胞凋亡影响;DNA倍体分析法检测HSJ对K562细胞周期时相影响。结果 HSJ对K562细胞增殖抑制率随剂量增加而增加,作用48 h半数抑制浓度(IC50)为1.35 mg/m L;1.0、2.0、4.0 mg/m L HSJ处理后,K562细胞凋亡率分别为(8.39±0.81)%、(8.88±1.12)%、(13.42±2.04)%,均高于对照组的(4.60±1.09)%(P<0.05);HSJ能够引起细胞周期进程的改变,与对照组比较,HSJ增加K562细胞中G2期细胞比例(P<0.05),降低S期时相细胞比例(P<0.05),降低增殖指数(PI)(P<0.05),HSJ将K562细胞阻滞于G2期。结论 HSJ对K562细胞具有增殖抑制作用,其机制与诱导细胞凋亡和阻滞K562细胞在G2期有关。
Objective To investigate the effects of aqueous extract of Acarya (GJ) on the apoptosis and cell cycle of human leukemia K562 cell line. Methods The inhibitory effect of HSJ on the proliferation of K562 cells was detected by MTT colorimetric assay. The effect of HSJ on the cell growth curve was observed by trypan blue exclusion staining. The apoptosis of K562 cells was detected by flow cytometry Effects of HSJ on the phase of K562 cells were analyzed by DNA ploidy assay. Results The inhibitory rate of HSJ on K562 cells increased with increasing dose, and the IC50 of 48 h was 1.35 mg / m L after HSJ treatment. The apoptotic rates of K562 cells treated with 1.0, 2.0 and 4.0 mg / m L of HSJ were (8.39 ± 0.81)%, (8.88 ± 1.12)% and (13.42 ± 2.04)%, respectively, which were significantly higher than those in the control group (4.60 ± 1.09)% (P <0.05). HSJ could induce cell cycle progression, Compared with control group, HSJ increased the proportion of G2 phase cells in K562 cells (P <0.05), decreased the proportion of phase cells in S phase (P <0.05) and decreased the proliferation index (PI) (P <0.05) G2 period. Conclusion HSJ can inhibit the proliferation of K562 cells. The mechanism is related to the induction of apoptosis and the arrest of K562 cells in G2 phase.
