目的探讨甘草甜素的抗肿瘤作用和机制。方法利用小鼠颌下腺纤维肉瘤细胞株,通过细胞生长状态检测、光电镜水平的形态学观察、细胞周期分析等方法,观察甘草甜素对纤维肉瘤细胞的增殖抑制作用。结果甘草甜素600mg/L以上浓度能抑制纤维肉瘤细胞的增殖,且随其浓度的增加,作用时间的延长,抑制效果越显著;甘草甜素可引起纤维肉瘤细胞形态学改变,可使纤维肉瘤细胞G1期向S期移行受阻。结论甘草甜素抑制纤维肉瘤细胞增殖的最低有效浓度为600mg/L以上;甘草甜素的抗肿瘤机制可能是通过干扰G1期细胞的某些代谢或/和阻碍DNA合成的启动来实现的。
Objective To investigate the anti-tumor effect and mechanism of glycyrrhizin. Methods The mouse submandibular gland fibrosarcoma cell line was used to observe the effect of glycyrrhizin on the proliferation of fibrosarcoma cells through the detection of cell growth status, morphological observation at the level of electro-optical microscope and cell cycle analysis. Results The concentration of glycyrrhizin 600 mg/L or more inhibited the proliferation of fibrosarcoma cells, and the inhibitory effect was more pronounced with the increase of its concentration and the prolonged action time. Glycyrrhizin could induce the morphological changes of fibrosarcoma cells and make fibrosarcoma possible. G1 phase migration to S phase is blocked. Conclusion Glycyrrhizin inhibits the proliferation of fibrosarcoma cells at a concentration of at least 600 mg/L. The anti-tumor mechanism of glycyrrhizin may be achieved by interfering with certain metabolism of G1 cells or/and inhibiting the initiation of DNA synthesis.
