建立了基质金属蛋白酶-1的活性测定方法及临床应用的部分意义。基质金属蛋白酶是广泛分布于细胞外基质等部位的蛋白酶类,目前已知有14种,基质金属蛋白酶-1主要水解作用于细胞外基质的Ⅰ,Ⅱ,Ⅲ型胶原蛋白,其最适底物为Ⅰ型胶原。该研究采用从胎牛皮肤分离提取Ⅰ型胶原蛋白,并经HPLC、电泳鉴定后,以3H-乙酸酐标记作为放射测定方法的底物,以此建立了基质金属蛋白酶-1活性的测定方法,并对部分临床和实验病例进行了测定意义研究,证实该方法操作可行,结果可靠,在实验性股骨头坏死及临床腰椎间盘退化病例中均获得与国外文献报告一致的结果。
Established the determination of MMP-1 activity and its clinical significance.
