目的探讨鹅绒委陵菜多糖(PAP)的抗肿瘤作用。方法采用MTT法研究PAP对体外培养人肝癌细胞SMMC-7721的增殖抑制作用;并将S180荷瘤小鼠随机分组,灌胃给予50,100,200 mg·kg-1剂量的PAP,研究PAP对荷瘤小鼠的抑瘤率和免疫功能的影响。结果体外实验结果显示PAP对SMMC-7721细胞的体外增殖有显著抑制作用,400 mg·L-1 PAP组抑制率最高,为43.57%;PAP对人肝癌SMMC-7721细胞的抑制作用呈剂量依赖性,拟合曲线呈线性相关。动物实验结果显示低、中、高3个剂量的PAP对S180荷瘤小鼠的抑瘤率分别为46.41%,52.29%和59.48%,其中环磷酰胺和低剂量的多糖组合抑瘤率与中、高剂量的多糖抑瘤率相当,并显著高于单用环磷酰胺组的抑瘤率;同时PAP能够显著提高T-淋巴细胞和巨噬细胞吞噬率。结论 PAP具有抗肿瘤活性和增强免疫的功能。
Aim To investigate the antitumor effect of polysaccharide from Potentilla. Methods The inhibitory effect of PAP on the proliferation of human hepatocellular carcinoma cell line SMMC-7721 was studied by MTT assay. S180 tumor-bearing mice were randomly divided into groups and were given orally with 50,100,200 mg · kg-1 PAP. The tumor inhibition rate and immune function. Results The results of in vitro experiments showed that PAP significantly inhibited the proliferation of SMMC-7721 cells in vitro, and the highest inhibitory rate was 43.57% in the 400 mg · L -1 PAP group. The inhibitory effect of PAP on human hepatocellular carcinoma SMMC-7721 cells in a dose-dependent manner , Fitting curve is linearly related. Animal experiments showed that low, medium and high doses of PAP on S180 tumor-bearing mice inhibition rates were 46.41%, 52.29% and 59.48%, respectively, of which cyclophosphamide and low dose of polysaccharide combination of tumor inhibition rate and , High dose of polysaccharide inhibition rate was quite high, and was significantly higher than the cyclophosphamide alone group tumor inhibition rate; while PAP can significantly increase the phagocytosis of T-lymphocytes and macrophages. Conclusions PAP has anti-tumor activity and enhances immune function.
