研究了水提-醇沉法提取紫山药糖蛋白。用正交设计试验考察提取温度、料液比、提取时间对提取率的影响。并分别用DEAE-52离子交换层析柱和Sephadex G-75葡聚糖凝胶柱对粗提物纯化。结果表明,提取条件为温度60℃,料液比1∶20(g/mL),提取时间3 h。粗提物经DEAE-52,Sephadex G-75柱层析纯化后得到1个糖蛋白组分,该糖蛋白经SDS-PAGE电泳分析,测得分子量为1.35×104 Da。
The extraction of purple yam glycoprotein by water extraction and alcohol precipitation was studied. Orthogonal design test was used to investigate the effects of extraction temperature, solid-liquid ratio and extraction time on extraction rate. The crude extracts were purified by DEAE-52 ion exchange chromatography and Sephadex G-75 sephadex. The results showed that the extraction conditions were temperature 60 ℃, solid-liquid ratio 1:20 (g / mL) and extraction time 3 h. The crude extract was purified by DEAE-52 and Sephadex G-75 column chromatography to obtain one glycoprotein. The glycoprotein was analyzed by SDS-PAGE and the molecular weight was 1.35 × 104 Da.
