目的研究柴胡皂苷d对肝星状细胞ERα和ERβ蛋白的调节作用。方法体外培养肝星状细胞系HSC-T6,细胞铺板24 h后,用柴胡皂苷d或雌二醇处理HSC-T6细胞;或者先以雌激素受体拮抗剂(ICI-182.780)预处理细胞30 min后,再用柴胡皂苷d或雌二醇处理细胞。用Western blot方法观察柴胡皂苷d对雌激素受体表达的影响。结果柴胡皂苷d(5μmol·L~(-1))和雌二醇(1μmol·L~(-1))处理细胞后能显著提高HSC-T6细胞ERα和ERβ蛋白的表达水平,且作用24 h后的上调作用最显著(P<0.01);柴胡皂苷d和雌二醇上调ERβ表达的作用比上调ERα表达的作用强。结论柴胡皂苷d能够上调HSC-T6细胞ERα和ERβ蛋白表达,是一种雌激素受体调节剂。
Objective To study the regulation effect of saikosaponin d on the expression of ERα and ERβ in hepatic stellate cells. Methods Hepatic stellate cell line HSC-T6 was cultured in vitro, HSC-T6 cells were treated with saikosaponin d or estradiol 24 hours after plating, or pretreated with estrogen receptor antagonist (ICI-182.780) After 30 min, the cells were treated with saikosaponin d or estradiol. The effect of saikosaponin d on estrogen receptor expression was observed by Western blot. Results Saikosaponin d (5μmol·L -1) and estradiol (1μmol·L -1) treatment significantly increased the expression of ERα and ERβ in HSC-T6 cells, and the effect of 24 (P <0.01). Upregulation of ERβexpression by saikosaponin d and estradiol was stronger than that of ERα upregulation. Conclusion Siji saponin d can up-regulate the expression of ERα and ERβ in HSC-T6 cells and is an estrogen receptor modulator.
