目的探讨RASSF1A基因对胃癌SGC7901细胞蛋白表达谱的影响。方法建立SGC7901和RASSF1A-SGC7901细胞双向凝胶电泳图谱,筛选和鉴定部分分离较好的差异蛋白质点。RT-PCR验证差异基因mRNA的表达。结果建立了分辨率较高、重复性较好的RASSF1A-SGC7901细胞和SGC7901细胞的二维凝胶电泳图谱。筛选出8个差异蛋白质点,包括galectin-1,TRP-14,ACBP,PSMB5,PSMB4,TIM,vimentin及CD79α。RASSF1A过表达诱导了胃癌细胞SGC7901的galectin-1、TRP-14、ABCP的mRNA表达上调。结论 galectin-1,TRP-14及ABCP可能参与了RASSF1A抑制胃癌细胞SGC7901的生长。
Objective To investigate the effect of RASSF1A gene on the protein expression profile of gastric cancer cell line SGC7901. Methods The two-dimensional gel electrophoresis pattern of SGC7901 and RASSF1A-SGC7901 cells was established to screen and identify differentially expressed protein spots. RT-PCR to verify the differential gene mRNA expression. Results Two-dimensional gel electrophoresis of RASSF1A-SGC7901 cells and SGC7901 cells with high resolution and good reproducibility was established. Eight differential protein spots were screened, including galectin-1, TRP-14, ACBP, PSMB5, PSMB4, TIM, vimentin and CD79α. RASSF1A overexpression induced up-regulation of galectin-1, TRP-14 and ABCP mRNA expression in gastric cancer SGC7901 cells. Conclusion galectin-1, TRP-14 and ABCP may be involved in the inhibition of RASSF1A on the growth of gastric cancer cell line SGC7901.
