用改进的Dowex-1×2短柱层析法分离底物3H-cAMP及产物3H-AMP测定了日本血吸虫匀浆中cAMP磷酸=酯酶的活力,结果表明血吸虫该酶的最适pH在7.5-8.5之间,虫令10天以上的血吸虫酶活力随虫令而上升,41天时酶活力约为104±3×10~3cpm/对/分,雄虫酶活力略高于雌虫。酶活力主要分布于虫体内,体表酶活力很低仅为匀浆中的1/30,在培养中亦不见释出。CTP及GTP对虫该酶有抑制作用。3.3mM敌百虫抑制酶活力65%,呋喃丙胺、硝唑咪及吡喹酮并不明显抑制虫该酶的活力。
The 3H-cAMP and 3H-AMP products were separated by modified Dowex-1 × 2 short column chromatography. The activity of cAMP phosphodiesterase in the homogenate of Schistosoma japonicum was determined. The results showed that the optimum pH of the enzyme was 7.5 -8.5, the insecticidal activity of Schistosoma japonicum increased more than 10 days, and the activity of 41 days was about 104 ± 3 × 10 ~ 3 cpm / pair / min. The activity of male enzyme was slightly higher than that of female. The enzyme activity is mainly distributed in the body of worms, the body surface enzyme activity is very low, only 1/30 of the homogenate, also not seen in the culture.
