本文用经典恒温动力学方法研究蝮蛇抗栓酶的热稳定性。测定了不同温度不同时间处理后酶失活变化的吸收度,计算出酶活力及酶失活的速度常数,按Arrhenius指数定律处理得回归方程式lnK=1.2839×10~4/T+33.97.由此计算出酶失活变化的活化能E=107.2±0.1KJmol~(-1).推算出室温下贮存期t_(6.9)~(20℃)=6.38Years.经方差分析该方法重现性好,结果准确。
In this paper, the classical thermostatic kinetic method is used to study the thermal stability of viper antithrombin. The enzyme activity and the rate constant of enzyme inactivation were calculated according to the changes of enzyme inactivation after different temperature treatments, and the regression equation lnK = 1.2839 × 10 ~ 4 / T + 33.97 according to the Arrhenius index law. The activation energy E = 107.2 ± 0.1KJmol -1 was calculated and the storage period t 6.9 ~ (20 ℃) = 6.38Years was calculated at room temperature.It was found that the method has good reproducibility by variance analysis, The result is accurate.
