本文研究了新的类抗心律失常药KW-3407,5-{(2-(diethylamino)ethyl]amino}-7-methoxy-5,11-dihydro [1]benzoxepino[3, 4,-b]pyridine1.5 fumarate,对犬实验性心律失常的影响以及对离体窦房结标本和乳头肌标本的影响,实验结果表明,静注KW-3407 20mg/kg,能够有效地对抗氟烷-肾上腺素所诱发的心律失常、洋地黄所诱发的心律失常以及冠状动脉二步结扎法所诱发的犬实验性室性心律失常(P<0.05或P<.01)。本文还通过测定不同时间的血药浓度,计算了能使心律失常的比值(单位时间内异位室性节律占总心跳次数的百分比)降低50%时的血药浓度IC_(50)。其结果,KW-3407抑
In this paper, a new antiarrhythmic drug KW-3407, 5-{(2-(diethylamino)ethyl]amino}-7-methoxy-5,11-dihydro [1]benzoxepino[3, 4,-b]pyridine1 was studied. .5 fumarate, effects on experimental arrhythmia in dogs and effects on isolated sinus node specimens and papillary muscle specimens. The experimental results show that intravenous injection of KW-3407 20 mg/kg can effectively combat halothane-adrenergic conditions. Induced arrhythmias, digitalis-induced arrhythmias, and canine experimental ventricular arrhythmias induced by two-step ligation of the coronary artery (P<0.05 or P<.01).This article also determined plasma concentrations at different times. We calculated the blood drug concentration IC_(50) that reduced the arrhythmia ratio (percentage of ectopic rhythm per unit time in the total heart rate per unit time) by 50%.
