This article describes the extraction of immunoglobulin G (IgG) from mammalian serum or ascites using a simple, two-step procedure. That is, albumin and other non-IgG proteins are first precipitated with caprylic acid, and then the IgG fraction is precipitated with ammonium sulfate. The authors believe that the key to this method is that if the caprylic acid is completely precipitated, the impurities will be reduced in the next ammonium sulfate precipitation of IgG. To this end, we must pay attention to the following factors: (1) Serum concentrations, serum dilution of 3 to 4 times, albumin and non-IgG protein precipitation complete, almost no precipitation in the IgG.
