目的探讨蕨麻(Pa)正丁醇提取物对大鼠心肌缺血再灌注损伤的保护作用。方法 Wistar大鼠60只随机分为6组:对照组(C组)、缺血组(I组)、缺血再灌注1 h组(R1h组)、缺血再灌注2 h组(R2h组)、蕨麻正丁醇提取物+缺血再灌注1 h组(Pa+R1h组)和蕨麻正丁醇提取物+缺血再灌注2 h组(Pa+R2h组)。放射免疫法测量各组血清内皮素-1(ET-1)水平;反转录聚合酶链式反应(RT-PCR)、蛋白质印迹(Western Blot)检测各组心肌组织中ET-1mRNA和蛋白的表达。结果与C组比较,I组的血清ET-1水平、心肌ET-1蛋白水平均明显升高,差异有统计学意义(P<0.05)。R1h组、R2h组的血清ET-1水平、心肌ET-1 mRNA和蛋白表达均高于C组和I组,差异有统计学意义(P<0.05)。Pa+R1h组和Pa+R2h组的血清ET-1水平、心肌ET-1 mRNA和蛋白表达与R1h组、R2h组比较明显降低,差异有统计学意义(P<0.05)。结论 ET-1在大鼠心肌缺血再灌注损伤过程中表达上调,蕨麻正丁醇提取物可降低ET-1的表达。
Objective To investigate the protective effect of n - Butanol extract of Pae on myocardial ischemia reperfusion injury in rats. Methods Sixty Wistar rats were randomly divided into 6 groups: control group (C group), ischemia group (I group), ischemia / reperfusion 1 h group (R1h group), ischemia / reperfusion 2h group , Butanol extracts of Phellodendron chinense + 1 h group of Ischemia reperfusion (Pa + R1h) and n-Butyl alcohol extract of Brachydanin + 2 h group of Ischemia reperfusion (Pa + R2h group). The levels of ET-1 in each group were measured by radioimmunoassay. The levels of ET-1 mRNA and protein in each group were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot expression. Results Compared with group C, serum ET-1 level and myocardial ET-1 protein level of group I were significantly increased, the difference was statistically significant (P <0.05). The levels of ET-1 and ET-1 mRNA and protein in R1h and R2h groups were significantly higher than those in C and I groups (P <0.05). The level of ET-1 in Pa + R1h group and Pa + R2h group was significantly lower than that in R1h group and R2h group (P <0.05). Conclusions ET-1 is up-regulated in the process of myocardial ischemia-reperfusion injury in rats, and n-butanol extract of Pterocarpus can reduce the expression of ET-1.
