本研究报道了裸露心肌标本的制备及研究肌钙蛋白Ca~(2+)敏感性的技术方法。10mmol/L EGTA裸露液浸浴大鼠右室乳头肌,其肌膜通透性明显增高,表现为去除舒张液中ATP和磷酸肌酸后,心肌立即挛缩。最适裸露时间为24h。此时最大Ca~(2+)激活张力(T_(max)最高;收缩和舒张至50%T_(max)所需时间T_(1/2)和R_(1/2)最短。张力-pCa(Ca~(2+)摩尔浓度的负对数)关系曲线是S形,且同一标本先后两次实验的张力-pCa关系及50%T_(max)时的pCa(pCa_(50))均无明显差别,而茶碱可使张力-pCa关系左移,pCa_(50)增高。表明高通透心肌标本功能稳定,且可反映肌钙蛋白Ca~(2+)敏感性的改变。
