目的探讨缺氧诱导因子-1α(HIF-1α)、HIF-2α、血管内皮生长因子(VEGF)和微血管密度(MVD)在肾透明细胞癌中的表达情况,分析四者的关系及意义。方法应用免疫组化方法分析107例肾透明细胞癌中HIF-1α、HIF-2α、VEGF和MVD的表达情况。结果本组HIF-2α的阳性染色率(80·5%)明显高于HIF-1α(69·2%)(P<0·01)。HIF-1α和HIF-2α阳性组中VEGF的阳性率(90·5%和89·8%)明显高于它们阴性组的VEGF阳性率(51·5%和26·3%),P<0·01。HIF-2α和VEGF的表达与MVD存在正相关关系,(Spearman相关系数分别为0·500和0·513),P<0·01。但肿瘤的病理分级和分期与HIF-1α、2α的表达无相关性,P>0·05。结论HIF-2α在肾透明细胞癌中的表达比HIF-1α更广泛,两者与VEGF的表达正相关。HIF-2α对VEGF的正向调控和促血管生成作用都明显强于HIF-1α。未发现两者的表达与肿瘤病理分级和分期的相关性。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α), HIF-2α, vascular endothelial growth factor (VEGF) and microvessel density (MVD) in renal clear cell carcinoma and to analyze their relationship and significance. Methods Immunohistochemistry was used to analyze the expression of HIF-1α, HIF-2α, VEGF and MVD in 107 cases of clear cell renal cell carcinoma. Results The positive staining rate of HIF-2α in this group (80 · 5%) was significantly higher than that of HIF-1α (69 · 2%) (P <0.01). The positive rates of VEGF (90.5% and 89.8%) in HIF-1α and HIF-2αpositive groups were significantly higher than those in their negative groups (51.5% and 26.3%, P <0 · 01. There was a positive correlation between the expression of HIF-2α and VEGF and MVD (Spearman’s correlation coefficients were 0.5 · 0 and 0 · 513 respectively), P <0.01. However, the pathological grading and staging of tumor had no correlation with the expression of HIF-1α and 2α (P> 0.05). Conclusions The expression of HIF-2α in renal clear cell carcinoma is more extensive than that of HIF-1α, both of them are positively correlated with the expression of VEGF. HIF-2α positive regulation of VEGF and angiogenesis were significantly stronger than HIF-1α. No correlation was found between the expression of these two proteins and tumor pathological grade and staging.
