目的:探讨特异性抑制NRP2基因表达对人胃癌细胞SGC7901裸鼠移植瘤生长及淋巴管新生的影响。方法:采用小RNA干扰方法,构建shNRP2质粒,稳定转染入SGC7901细胞株,Westen blot检测转染前后NRP2蛋白的表达。建立人胃癌细胞SGC7901裸鼠移植瘤模型,随机分为shNRP2组(实验组)、shCon组(HK阴性对照组)和正常对照组,观察移植瘤的生长情况。6周后处死裸鼠,免疫组化检测NRP2蛋白的表达及微淋巴管密度(Micro-vessel density,MLD)。结果:成功构建shNRP2质粒,与另两组比较,shNRP2组细胞NRP2蛋白表达明显降低,且移植瘤组织生长、NRP2表达及MLD明显受抑制。结论:抑制NRP2基因的表达,可以抑制胃癌裸鼠移植瘤的生长及淋巴管的形成,NRP2基因有可能成为一个潜在的胃癌生物治疗靶点。
Objective: To investigate the effects of NRP2 gene silencing on the growth and lymphangiogenesis of human gastric cancer SGC7901 xenografts in nude mice. Methods: The shNRP2 plasmid was constructed by small RNA interference and stably transfected into SGC7901 cell line. The expression of NRP2 protein was detected by Westen blot. SGC7901 human gastric cancer xenografts model was established and randomly divided into shNRP2 group (experimental group), shCon group (HK negative control group) and normal control group to observe the growth of xenograft tumors. After 6 weeks, the nude mice were sacrificed and the expression of NRP2 protein and micro-vessel density (MLD) were detected by immunohistochemistry. Results: The shNRP2 plasmid was successfully constructed. Compared with the other two groups, the expression of NRP2 protein in the shNRP2 group was significantly decreased, and the growth of the tumor tissue, NRP2 expression and MLD were significantly inhibited. Conclusion: Inhibition of NRP2 gene expression can inhibit the growth and lymphangiogenesis of gastric cancer xenografts in nude mice. NRP2 gene may be a potential therapeutic target for gastric cancer.
