目的探讨胞嘧啶脱氨酶(CD)/5-氟胞嘧啶(5-FC)基因治疗联合放疗对裸鼠宫颈癌移植瘤的生长抑制作用,以及CD/5-FC基因治疗与放疗之间是否具有协同作用。方法将24只鼠龄6~8周成功接种了HeLa细胞的BALB/C雌性裸鼠,随机分为对照组、基因治疗组、放疗组以及联合治疗组,每组6只,观察各组治疗前后的肿瘤体积、肿瘤生长抑制率及肿瘤体积增加倍数。结果(1)治疗21d后,基因治疗组裸鼠的肿瘤体积是(728±201)mm3,联合治疗组是(357±113)mm3,放疗组是(739±419)mm3,对照组是(1168±380)mm3,基因治疗组、联合治疗组、放疗组肿瘤体积均小于对照组,3组分别与对照组比较,差异有统计学意义(P<0.05);联合治疗组肿瘤体积小于基因治疗组及放疗组,前者分别与后两组比较,差异有统计学意义(P<0.05)。(2)放疗组的肿瘤生长抑制率为36.74%,基因治疗组为37.66%,联合治疗组为69.45%,联合治疗组明显高于放疗组及基因治疗组,前者分别与后两组比较,差异有统计学意义(P<0.05)。(3)联合治疗组平均肿瘤体积增加倍数为7.71倍,基因治疗组为19.16倍,放疗组为18.86倍,对照组为40.57倍。析因设计的方差分析结果,CD/5-FC基因治疗与放疗之间有交互作用(P=0.03),当与放疗联合使用时,CD/5-FC基因治疗的疗效大于其单独使用时所产生的疗效;当与CD/5-FC基因治疗联合应用时,放疗的疗效大于其单独使用时所产生的疗效。结论CD/5-FC基因治疗与放疗联合应用,对裸鼠宫颈癌移植瘤的生长具有抑制作用,并且两者之间具有协同作用。
