目的 :观察急性心肌梗死后心室结构的主要变化及心肌局部血管紧张素 (Ang )和羟脯氨酸 (HP)含量的变化 ,并观察黄芪的治疗作用。方法 :全部实验用兔 6 0只 ,分 3组 ,第 1组 2 0只作对照 ,第 2组 2 0只制备心肌梗死模型 ,第 3组 2 0只用黄芪治疗。于术后第 6周处死动物 ,用精确天平及精密卡尺直接测量左室重量、全心重量及梗死区、游离壁和室间隔厚度。采集标本 ,检测梗死区、游离壁及正常兔心肌 Ang 和 HP含量。结果 :梗死后全心重量及左室重量均增加 ,差异显著 ,室间隔及游离壁厚度增加 ,梗死区室壁变薄 ,差异显著 ;心肌梗死后心肌局部Ang 含量显著增加 ;梗死区 HP含量显著增加 ,而游离壁的含量略有增加 ;经黄芪治疗后左室重量和全心重量均下降 ,其中全心重量尚未恢复正常 ;心肌 Ang 含量明显下降 ,梗死区 HP含量略有下降。结论 :心肌梗死后心室构型发生明显变化 ,Ang 和 HP参与左室重构 ,黄芪有一定的治疗作用。
OBJECTIVE: To observe the changes of ventricular structure and the content of angiotensin (Ang) and hydroxyproline (HP) in myocardium after acute myocardial infarction (AMI) and to observe the therapeutic effect of Astragalus membranaceus. Methods: Sixty rabbits were divided into three groups, 20 rabbits in the first group were used as controls, 20 rabbits in the second group were given myocardial infarction model, and 30 rabbits in the third group were treated with Astragalus membranaceus. Animals were sacrificed at 6 weeks after surgery. Left ventricular mass, total heart weight and infarct size, free wall and interventricular septum thickness were measured directly with a precision balance and a precision caliper. Specimens were collected to detect the contents of Ang and HP in infarcted area, free wall and normal rabbit myocardium. Results: After the infarction, the total heart weight and left ventricular weight increased significantly, the ventricular septum and the free wall thickness increased, the infarct wall thinning, the difference was significant; myocardial Angiotensin II was significantly increased after myocardial infarction; While the content of free wall increased slightly. The weight of left ventricle and whole heart weight decreased after treatment with Astragalus membranaceus, the whole heart weight had not returned to normal, the content of Angiotensin Ⅱ in myocardium decreased obviously, and the content of HP in infarction area decreased slightly. Conclusion: The changes of ventricular configuration after myocardial infarction have obvious changes. Ang and HP are involved in left ventricular remodeling. Astragalus has a certain therapeutic effect.
