目的探讨Survivin反义寡核苷酸及VP-16对K562细胞株凋亡的影响。方法将Survivin反义寡核苷酸通过脂质体转染K562细胞株,用免疫组化法测定转染前后K562细胞survivin基因及Caspase-3的表达差异,并加入足叶乙甙,用Tenu l及Annexin V-FITC测定各组细胞的凋亡率。结果转染后survivin基因表达下降,Caspase-3的表达升高,K562细胞生长受抑,细胞凋亡率升高;联合反义寡核苷酸与足叶乙甙,细胞凋亡率明显增加。结论Survivin反义寡核苷酸能够促进白血病细胞株K562的凋亡,能够提高K562对足叶乙甙的敏感性。
Objective To investigate the effect of Survivin antisense oligonucleotide and VP-16 on the apoptosis of K562 cell line. Methods Survivin antisense oligonucleotide was transfected into K562 cells by lipofectamine 2000. The expression of survivin and Caspase-3 in K562 cells was detected by immunohistochemistry. The cells were treated with Tenu l And Annexin V-FITC to determine the apoptosis rate of each group. Results After transfection, the expression of survivin gene was decreased, the expression of Caspase-3 was increased, the growth of K562 cells was inhibited and the apoptosis rate was increased. Combined with antisense oligodeoxynucleotides and etoposide, the apoptosis rate was significantly increased. Conclusion Survivin antisense oligonucleotide can promote the apoptosis of leukemia cell line K562 and increase the sensitivity of K562 to etoposide.
