目的:以带有球茎的不定芽为外植体,探讨不同植物生长物质与活性炭(AC)对鼎湖山紫背天葵组培苗生根和移栽的影响。方法:组织培养单因子试验法。结果:NAA 0.3 mg/L+IBA 0.2 mg/L的组合较好地诱导球茎芽丛生根,但根数仍较少,根细、短小,幼小的不定芽萌发数量较多。添加300 mg/L活性炭明显提高生根质量并且抑制细芽点的分化,根数多达15.5条,根长范围在2.0~5.1 cm,根系发达,并且组培苗植株较高、球茎和叶片质量佳,生长健壮。将生根的组培苗带球茎分散为丛芽的形式移栽到泥炭土-珍珠岩(3∶1)的混合基质中,移栽后成活率达到100%,植株出苗快,长势旺盛。结论:以MS+蔗糖30 g/L+NAA 0.3 mg/L+IBA 0.2 mg/L+活性炭300mg/L+卡拉胶7.0 g/L作为紫背天葵组培苗的生根体系,是离体快繁和保存本地特色植物的有效途径。
OBJECTIVE: To investigate the effects of different plant growth materials and activated carbon (AC) on the rooting and transplanting of tissue culture seedlings of Dinghushan Purpurea in vitro using adventitious buds with bulbs as explants. Methods: Tissue culture single factor test method. Results: The combinations of NAA 0.3 mg / L and IBA 0.2 mg / L could induce the bulbous shoots to root well, but the number of roots was still small, and the number of adventitious buds was small. Addition of 300 mg / L activated charcoal significantly increased rooting quality and inhibited the differentiation of fine buds, with the number of roots reaching 15.5 and the root length ranging from 2.0 cm to 5.1 cm. The roots were well developed and the tissue culture plants had higher plantlets and better quality of bulbs and leaves , Grow robust. The rooted tissue culture seedling bulbs were transplanted into the mixed matrix of peat soil-perlite (3: 1) in the form of cluster buds. The survival rate after transplanting reached 100%. The plants emerged quickly and grew vigorously. Conclusion: MS + sucrose 30 g / L + NAA 0.3 mg / L + IBA 0.2 mg / L + activated carbon 300 mg / L + carrageenan 7.0 g / L was the root system of tissue culture seedlings of Prunus humilis An effective way of local characteristic plants.
