目的:观察生脉注射液对脂多糖诱导的脓毒症大鼠炎症反应和血管内皮细胞损伤的影响。方法:将雄性Sprague-Dawley(SD)大鼠54只,随机分为对照组,脂多糖组和生脉治疗组,每组各18只。对照组腹腔注射生理盐水,脂多糖组和生脉治疗组腹腔注射脂多糖制备脓毒症大鼠模型,生脉治疗组实验前24h,注射脂多糖前1h分别静脉注射生脉注射液2ml/kg,制模成功后抽血检测血清白介素6(IL-6)和肿瘤坏死因子α(TNF-α),血清血管假性血友病因子(vWF)及血栓调节蛋白(TM)水平。结果:脂多糖组和生脉治疗组血清中IL-6、TNF-α、vWF和TM明显高于对照组(P<0.05);生脉治疗组IL-6、TNF-α、vWF、TM浓度较脂多糖组明显降低(P<0.05)。结论:生脉注射液可抑制脓毒症炎性因子的释放,对于脓毒症血管内皮细胞损伤具有一定的保护作用。
Objective: To observe the effect of Shengmai injection on inflammatory reaction and injury of vascular endothelial cells induced by lipopolysaccharide in septic rats. Methods: 54 male Sprague-Dawley (SD) rats were randomly divided into control group, lipopolysaccharide group and Shengmai treatment group, with 18 rats in each group. The rats in control group were injected intraperitoneally with saline, LPS group and Shengmai treatment group, respectively. Lipopolysaccharide was injected intraperitoneally to prepare rats model of sepsis. Shengmai treatment group was injected intraperitoneally with Shengmai injection 2ml / kg Blood samples were collected for determination of serum levels of interleukin 6 (IL-6), tumor necrosis factor-α (TNF-α), vWF, and thrombomodulin (TM). Results: The levels of IL-6, TNF-α, vWF and TM in serum of LPS group and Shengmai treatment group were significantly higher than those of control group (P <0.05) Compared with lipopolysaccharide group was significantly lower (P <0.05). Conclusion: Shengmai injection can inhibit the release of inflammatory cytokines in sepsis, which has a protective effect on vascular endothelial cell injury in sepsis.
