目前国内外对油菜菌核病抗(耐)病性的鉴定和筛选,大多依据自然发病或大田人工诱发病圃的抗性表现,受环境影响大、鉴定标准难于掌握,且结果不稳定。本文介绍经作者接种油菜叶片根据病斑大小区分抗(耐)、感性的快速方法。 菌源制备:取田间病株上收集的油菜菌核病菌核,消毒后接种于PDA平皿培养基上,25℃条件下黑暗培养3-5天,再挑取边缘生长的菌丝块接种于液体培养基上,每升培养基含368mgMgSO_47H_2O,149mg KCl,1.00gNH_4NO_3,680mgKH_2PO_4,11.3mg ZnSO_4 7H_2O,13.9mg MgSO_4 H_2O,6.6mgFeCl_3 6.25mgCuSO_4 5H_2O以及0.5g酵母浸提物、73.7mM D-葡萄糖和56mM琥珀酸钠。接种后振荡培养5-6天,当菌丝体呈灰白色胶质状时,转移至消毒的麦粒培养基上扩大繁殖作为供试菌源。 接种:在油菜盛花期,随机选取待鉴定油菜品种(系)植株中下部同一叶位生长正常的叶5-10片,每叶接种菌源麦粒5-6颗于叶片不同部位,同时套上装有1个保湿用湿棉球
