目的改进大葱减数分裂观察的实验教学;观察秋水仙碱(COM)对大葱减数分裂的影响及可能的机制。方法采用不同COM浓度和不同COM处理时间来观察减数分裂指数(MeI)的变化。结果COM的浓度在50μg/ml,处理时间为24小时左右时,MeI值最高;COM与微管的结合是可逆的,并且可以影响细胞DNA的合成。结论COM可以有效地影响大葱减数分裂并使MeI值显著增大;同时也使大葱减数分裂观察的实验教学得以改进。
Objective To improve the experimental teaching of observation of meiosis of Welsh onion, and to observe the effect of colchicine (COM) on meiosis of Welsh onion and its possible mechanism. Methods The meiosis index (MeI) was observed using different COM concentrations and different COM treatment times. Results When the concentration of COM was 50μg/ml and the treatment time was about 24 hours, the MeI value was the highest. The combination of COM and microtubules was reversible and could affect the DNA synthesis of cells. Conclusion COM can effectively affect the meiosis of Welsh onion and increase the MeI value significantly. At the same time, it also improves the experimental teaching of meiotic observation of Welsh onion.
