常规培养皿和富含氨基酸的培养液存在较强的拉曼吸收,为了获得细胞的拉曼光谱,我们对其前处理后进行拉曼光谱检测。目的:排除来自培养环境的干扰,检测细胞的拉曼光谱。方法:①漂洗细胞以排除培养液干扰;②固定细胞以保证其结构完整;③使细胞脱水以排除水的干扰;④富集细胞成团,使其具有一定高度,降低来自于背底(培养皿)的干扰。实验结果显示:前处理有效排除了来自培养环境的干扰,在532 nm和785 nm激发光源下均检测到了仅来自于细胞的拉曼光谱。
