目的:为探讨宁波市不同地区、不同年份、不同来源的霍乱病原菌之间的分子分型和遗传相关性,研究宁波市霍乱分子流行病学特点。方法:采用限制性内切酶NotⅠ对2004~2006年宁波地区霍乱疫情中分离的52株O139群霍乱弧菌进行脉冲场凝胶(PFGE)分子分型研究。结果:52株O139群霍乱弧菌基因组DNA经NotⅠ酶酶切、PFGE图谱分析,DNA条带得到有效分离,在20~500 KB区间产生约20条带。所有菌株按条带数量和位置的不同分为5个型别。结论:PFGE对来自不同地区、不同年份、不同来源的霍乱弧菌遗传相关性有较好的分辨能力。
OBJECTIVE: To investigate the molecular typing and genetic correlation of cholera pathogens in different regions, different years and different sources in Ningbo and to study the epidemiological characteristics of cholera in Ningbo. Methods: Fifty-two strains of Vibrio cholerae O139 isolates from 2004 to 2006 in Ningbo area were genotyped by pulsed-field gel electrophoresis (PFGE) using restriction endonuclease NotⅠ. Results: 52 strains of V. cholerae O139 genomic DNA were digested by Not Ⅰ enzyme and analyzed by PFGE. DNA bands were effectively separated and about 20 bands were produced in the range of 20 ~ 500 KB. All strains according to the number and location of the band is divided into five types. Conclusion: PFGE has better discrimination ability to Vibrio cholerae from different regions, different years and different sources.
