目的检测透骨草提取物对人卵巢癌SKOV-3细胞增殖的影响,探讨透骨草提取物影响SKOV-3细胞增殖的相关分子机制。方法采用倒置显微镜观察透骨草提取物对SKOV-3细胞增殖的影响,采用免疫组织化学方法检测SKOV-3细胞P53蛋白和c-Myc蛋白的表达。结果倒置显微镜观察可见,37.8μg/mL透骨草提取物处理的SKOV-3细胞皱缩变形,轮廓不规则、不清晰,细胞脱壁增加。37.8μg/mL透骨草提取物处理的SKOV-3细胞P53蛋白和c-Myc蛋白的表达明显低于对照组(P<0.05)。结论透骨草提取物可以显著抑制SKOV-3细胞的增殖,其机制可能与下调P53蛋白和c-Myc蛋白表达有关。
Objective To detect the effect of the extract of Huutongcao on the proliferation of human ovarian cancer SKOV-3 cells and to explore the molecular mechanism of the effect of the extract of Huutongcao on the proliferation of SKOV-3 cells. Methods Inverted microscope was used to observe the effect of the extract of SKOV-3 on the proliferation of SKOV-3 cells. The expression of P53 protein and c-Myc protein in SKOV-3 cells was detected by immunohistochemistry. Results Inverted microscope showed that the SKOV-3 cells treated with 37.8μg / mL extract of Anemarrhena astigmatism had wrinkles, irregular contour, unclear appearance and increased cell wall detachment. The expression of P53 protein and c-Myc protein in SKOV-3 cells treated with 37.8μg / mL Ostrinia furnacalis was significantly lower than that in the control group (P <0.05). Conclusion The extract of Tougu grass can significantly inhibit the proliferation of SKOV-3 cells, and its mechanism may be related to down-regulation of P53 protein and c-Myc protein expression.
