目的:探讨金匮肾气丸对庆大霉素造成药物性耳聋豚鼠耳蜗螺旋神经节神经修复相关蛋白表达的影响。方法:30只豚鼠随机分成3组各10只,即正常组、模型组、治疗组。将模型组豚鼠按100mg/kg/d注射庆大霉素,治疗组在注射庆大霉素后灌服金匮肾气丸水煎剂,实验结束后取耳蜗标本,免疫组化观察其生长相关蛋白(GAP-43)及神经生长因子(NGF)的表达。结果:GAP-43、NGF在实验动物耳蜗螺旋神经节细胞中都有一定的表达;正常组表达较弱较少,其它各组表达均较正常组表达增加,其中治疗组表达高于模型组(P<0.01)。结论:金匮肾气丸可提高庆大霉素致聋豚鼠耳蜗GAP-43、NGF表达,对螺旋神经节细胞进行修复。
Objective: To investigate the effect of Jinkuishenqi Pills on the protein expression of cochlear spiral ganglion nerve repair induced by gentamycin in guinea pigs with drug-induced deafness. Methods: Thirty guinea pigs were randomly divided into 3 groups of 10, namely normal group, model group and treatment group. The model group of guinea pigs were injected gentamicin at 100mg / kg / d. The treatment group was given Jingui Shenqi Decoction after gentamicin injection. At the end of the experiment, cochlear samples were taken and the growth was observed by immunohistochemistry Protein (GAP-43) and nerve growth factor (NGF) expression. Results: GAP-43 and NGF were all expressed in the spiral ganglion cells of the experimental animals. The expressions of GAP-43 and NGF in the spiral ganglion cells of the experimental animals were weaker than those in the untreated group. The expressions of GAP-43 and NGF in the normal group were weaker than those in the normal group P <0.01). Conclusion: Jinkuishenqi Pill can increase the expression of GAP-43 and NGF in gentamicin-induced deafness guinea pig cochlea and repair spiral ganglion cells.
