                                目的:观察甲基强的松龙对局灶性脑缺血模型纹状体内TNFα、NO含量的影响。方法:用线栓法制备局灶性脑缺血模型,用ELISA方法检测TNFα,采用Griess试剂测定微透析液中NO的代谢产物亚硝酸盐含量。结果:缺血1h再灌注6h,缺血侧纹状体内NO及TNFα含量升高,给予3mg·kg-1甲基强的松龙,可以使两种物质的含量下降。结论:给予小剂量的甲基强的松龙(3mg·kg-1)可以降低脑缺血组织的TNFα、NO含量,可能对脑缺血损伤产生保护作用。
Objective: To observe the effect of methylprednisolone on the content of TNFα and NO in striatum of focal cerebral ischemia model. Methods: Focal cerebral ischemia model was established by thread occlusion method. TNFα was detected by ELISA. The nitrite content of NO metabolites in microdialysis fluid was determined by Griess reagent. Results: After ischemia for 1h and reperfusion for 6h, the content of NO and TNFα in ischemic lateral striatum increased. Methylprednisolone given at 3 mg · kg-1 decreased the content of both substances. CONCLUSION: Small doses of methylprednisolone (3 mg · kg -1) can reduce the levels of TNFα and NO in cerebral ischemic tissue and may have a protective effect on cerebral ischemia.
