放射治疗已成为治疗肿瘤的一种重要方法,但在治疗同时,高剂量的电离辐射会对机体各个组织器官造成损伤。为探讨高剂量电离辐射对恒河猴(Macaca mulatta)心、肝、脾、肺、肾及肠道组织的病理性损伤及相关细胞因子表达的影响,本研究用辐射能量9.5 Gy的60Coγ对3只雄性恒河猴照射4 h,于辐射后15 d处死并采集相关器官组织,采用苏木精-伊红染色(hematoxylin-eosin staining,HE)观察心、肝、脾、肺、肾及肠道组织病理损伤情况,同时采用免疫组化和图像分析技术检测各组织内肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白介素-6(interleukin-6,IL-6)、IL-10、IL-2和转化生长因子β1(transforming growth factorβ1,TGF-β1)的表达。结果表明,辐射后15 d,跟对照相比,恒河猴心、肝、脾、肺及肾组织发生明显病理变化,而且TNF-α、IL-6、IL-10、IL-2和TGF-β1表达量显著高于对照组(P<0.05),说明电离辐射所致的放射性损伤对相关细胞因子表达有影响,研究结果可为放射性损伤的诊断提供辅助参考。
Radiation therapy has become an important method of treatment of cancer, but at the same time, high doses of ionizing radiation damage the various tissues and organs of the body. To investigate the effect of high dose ionizing radiation on the pathological injury of heart, liver, spleen, lung, kidney and intestine and the expression of related cytokines in Macaca mulatta, Male Rhesus macaques were irradiated for 4 h, sacrificed 15 days after irradiation, and related organs were harvested. Heart, liver, spleen, lung, kidney and intestine were observed by hematoxylin-eosin staining Meanwhile, the expression of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and IL-6 in each tissue were detected by immunohistochemistry and image analysis. 10, IL-2 and transforming growth factor-β1 (TGF-β1). The results showed that the pathological changes of heart, liver, spleen, lung and kidney of rhesus monkey were observed at 15 days after irradiation, and the levels of TNF-α, IL-6, IL- β1 expression was significantly higher than that of the control group (P <0.05), indicating that radiation damage caused by ionizing radiation has an impact on the expression of related cytokines, and the results can provide reference for the diagnosis of radiation injury.
