目的:探讨雷公藤诱导的急性肾损伤大鼠模型中肾脏热休克蛋白70(HSP70)的分布及表达规律。方法:150只大鼠随机分为对照组(A)和低剂量组B(雷公藤20mg/(kg·d))以及高剂量组C(雷公藤40mg/(kg·d))一次性灌胃,将对照组和实验组分别于给药后6、12、24、36、48小时各取10只大鼠剖腹取肾,光镜、电镜下观察肾脏的病理变化,通过免疫组化法观察HSP70的表达情况。结果:HSP70在肾皮质肾小球周围近曲小管上皮细胞胞浆内分布增加,其表达量在给药后6h已经升高,12h达到高峰,48h仍高于正常对照组。同一时相点高剂量组较低剂量组HSP70的表达量高(P<0.01)。结论:中药雷公藤一次性大剂量给药即可引起急性肾损伤,在损伤的早期HSP70的表达亦显著增加,提示肾毒性药物早期即可引起肾小管上皮细胞的应激反应,可能与细胞的自我保护机制启动有关。
Objective: To investigate the distribution and expression of heat shock protein 70 (HSP70) in kidney of a rat model of acute kidney injury induced by Tripterygium wilfordii. Methods: 150 rats were randomly divided into control group (A) and low-dose group B (TWP<20mg/(kg·d)) and high-dose group C (TWP<40mg/(kg·d)) The rats of the control group and the experimental group were taken at 6th, 12th, 24th, 36th, and 48th hours after administration respectively to take a kidney laparotomy. The pathological changes of the kidney were observed under light microscope and electron microscope, and the HSP70 was observed by immunohistochemistry. The expression of the situation. RESULTS: The distribution of HSP70 in the cytoplasm of proximal tubular epithelial cells around the renal cortex glomeruli increased. The expression level of HSP70 increased at 6 h after administration, reached a peak at 12 h, and was still higher than that of the normal control group at 48 h. The expression of HSP70 in the high-dose group at the same time point was higher (P<0.01). Conclusion: Acute kidney injury can be caused by high-dose one-time administration of Tripterygium wilfordii. The expression of HSP70 is also significantly increased in the early stage of injury, suggesting that early renal toxicity can cause renal tubular epithelial cell stress response, possibly related to cellular The self-protection mechanism started.
