目的 研究单层培养不同胎龄人胎甲状旁腺细胞的形态及功能。方法 将 2 0周至 2 8周水囊引产的胎儿按周龄分为 3组 ,用胰酶消化法将胎儿甲状旁腺分离制成单细胞并培养 ,每日观察并记录甲状旁腺细胞的形态 ,每隔 48h留取甲状旁腺细胞培养的上清液 ,测定上清液中甲状旁腺激素 (PTH)的浓度。结果 形态学观察发现胎儿甲状旁腺细胞培养 48h后贴壁 ,细胞结构完整 ,群聚生长 ,但无明显细胞延展生长现象。第 11天以后细胞轮廓增强 ,胞质内可见颗粒状物 ,并可见悬浮的细胞碎片。上清液中PTH水平第 3、第 11天与其它各天均有显著性差异(P <0 .0 1) ;3个胎龄组之间PTH水平差异无显著性 (P >0 .0 1)。结论 胎儿甲状旁腺细胞培养至第 3天形态及分泌功能最佳 ;2 0周到 2 8周内周龄对甲状旁腺细胞的分泌无影响。
Objective To study the morphology and function of parathyroid cells cultured in monolayer with different gestational age. Methods Twenty-seven weeks old fetuses were divided into 3 groups according to the age of fetus. The fetuses were isolated and cultured into single cells by trypsin digestion. The morphology of parathyroid cells was observed and recorded daily. Parathyroid cell culture supernatants were collected every 48h, and the concentration of parathyroid hormone (PTH) in the supernatant was measured. Results Morphological observation showed that fetal parathyroid cells adherent after 48h culture, cell structure integrity, clustered growth, but no significant cell growth phenomenon. After the 11th day, the outline of the cells is enhanced. Granules can be seen in the cytoplasm and the suspended cell debris can be seen. PTH levels in supernatants were significantly different from other days on the 3rd and 11th days (P <0.01). There was no significant difference in PTH levels between the three groups (P> 0.05) ). Conclusion Fetal parathyroid cells have the best morphological and secretory function on the third day of culture, and have no effect on the secretion of parathyroid cells from 20 weeks to 28 weeks.
