目的以外源性腺苷前质——硫酸腺嘌呤注入大鼠,通过琥珀酸脱氢酶(SDH)电镜细胞化学的变化,探讨预处理效应。方法实验采用体重为200~300g雄性SD大鼠24只,分4组,每组6只,即正常对照组(Ⅰ组);缺血再灌组(Ⅱ组);缺血预处理组(Ⅲ组,按经典的Murry法);硫酸腺嘌呤预处理组(Ⅳ组)。按Nitro-BT法显色制作SDH光镜标本,按小川的亚铁氰化铜法制作电镜标本。光镜观察SDH活性参照Niles法分为4级。结果Ⅰ组SDH活性和心肌结构相对正常;Ⅱ组SDH活性降低明显,线粒体损伤严重;Ⅳ组近于Ⅰ组;Ⅲ组界于Ⅰ、Ⅳ组之间。结论外源性硫酸腺嘌呤注入大鼠具有与经典缺血预处理相类似的抗心肌缺血再灌损伤的效应
Objective To investigate the preconditioning effect of exogenous adenosine preconditioning-adenosine sulphate into rat by electron microscopy of succinate dehydrogenase (SDH). Methods Twenty-four male Sprague-Dawley rats weighing 200-300g were divided into 4 groups (6 rats in each group), namely normal control group (group Ⅰ), ischemia reperfusion group (group Ⅱ), ischemic preconditioning group (Ⅲ) Group, according to the classic Murry method); adenine sulfate pretreatment group (group Ⅳ). According to Nitro-BT method for the production of SDH light microscope specimens, according to Ogawa ferrocyanide production of electron microscopy specimens. Light microscopy SDH activity according to Niles method is divided into 4 levels. Results SDH activity and myocardial structure in group Ⅰ were relatively normal. SDH activity in group Ⅱ was significantly decreased and mitochondria were damaged severely; group Ⅳ was more than group Ⅰ; group Ⅲ was in group Ⅰ and Ⅳ. Conclusion The exogenous adenosine sulphate injected into rats has the similar effect of anti-ischemic and reperfusion injury as the classic ischemic preconditioning
