目的通过检测盐酸小檗碱单用或联合抗菌药物对表皮葡萄球菌(Staphylococcus epidermidis,Se.)形成生物被膜及所需基因的抑制作用。方法体外构建表皮葡萄球菌生物被膜模型,盐酸小檗碱单用或联合万古霉素、环丙沙星、红霉素分别干预生物被膜的形成,采用CRA平板法检测药物对Se.生物被膜的抑制作用,荧光定量RT-PCR方法检测药物作用前后表皮葡萄球菌icaA基因和agr基因的表达变化。结果盐酸小檗碱对icaA基因和agr基因的影响作用不及万古霉素、环丙沙星、红霉素;当不同浓度的盐酸小檗碱分别与万古霉素、环丙沙星、红霉素联用时,随着盐酸小檗碱的浓度增加,联合用药对icaA基因和agr基因的抑制作用逐渐减弱。结论中药单体(盐酸小檗碱)对Se.形成生物被膜及所需关键基因的表达有一定的抑制作用,但抑制作用不及抗生素(万古霉素、环丙沙星、红霉素),并且不能与抗生素(万古霉素、环丙沙星、红霉素)联合用药,它们之间存在拮抗作用。
OBJECTIVE: To determine the inhibitory effect of berberine hydrochloride alone or in combination with antibiotics on the formation of biofilms and desired genes in Staphylococcus epidermidis (Se.). Methods The biofilm model of Staphylococcus epidermidis was constructed in vitro. Berberine hydrochloride alone or in combination with vancomycin, ciprofloxacin and erythromycin were used to interfere with the formation of biofilms. The inhibition of Se biofilms by drugs was detected by CRA plate method. The role of fluorogenic quantitative RT-PCR method was used to detect the expression of icaA gene and agr gene in Staphylococcus epidermidis before and after drug treatment. Results Berberine hydrochloride had less effect on icaA gene and agr gene than vancomycin, ciprofloxacin and erythromycin; when different concentrations of berberine hydrochloride were compared with vancomycin, ciprofloxacin and erythromycin, respectively In combination, as the concentration of berberine hydrochloride increases, the inhibitory effect of combination therapy on icaA gene and agr gene gradually decreases. Conclusion The Chinese herbal monomer (berberine hydrochloride) has a certain inhibitory effect on the formation of biofilms of Se. and the expression of key genes, but the inhibitory effect is lower than that of antibiotics (vancomycin, ciprofloxacin, erythromycin), and Can not be combined with antibiotics (vancomycin, ciprofloxacin, erythromycin) combination of drugs, there is an antagonistic effect between them.
