芹菜素电化学探针与核酸作用机理的研究

通过电化学法研究了fsDNA影响下芹菜素(API)在金电极上的电极行为,优化确定了fsDNA的检测条件。结果表明,在4.0×10-7~7.0×10-6g/mL浓度范围内峰电流的增强与加入的fsDNA的浓度呈线性关系,线性方程为ΔIp/(μA)=3.20CfsDNA(10-5g/mL)+1.06,检测限为5.8×10-8g/mL,对模拟样品的测定结果令人满意;研究还表明芹菜素发生了3e传递行为,并有质子参与,且该行为受到扩散控制。通过循环伏安、紫外-可见光谱等方法对API与fsDNA间的作用机制进行了初步探讨,结果表明两者之间存在着静电作用,结合常数K=1.8×104L/mol。 The electrode behavior of apigenin (api) on gold electrode was studied by electrochemical method and the detection conditions of fsDNA were optimized. The results showed that there was a linear relationship between the increase of peak current and the concentration of fsDNA in the concentration range of 4.0 × 10-7 ~ 7.0 × 10-6g / mL, the linear equation was ΔIp / (μA) = 3.20CfsDNA (10-5g / mL) +1.06 with a detection limit of 5.8 × 10-8g / mL. The results of the determination of simulated samples are satisfactory. The study also shows that 3e transfer of apigenin takes place and proton is involved, and this behavior is controlled by diffusion. The mechanism of interaction between API and fsDNA was studied by cyclic voltammetry and ultraviolet-visible spectroscopy. The results showed that there was an electrostatic interaction between the two and the binding constant was K = 1.8 × 104L / mol.
