目的探讨啤酒花总黄酮对硫代乙酰胺所致的小鼠急性肝损伤的保护作用。方法采用硫代乙酰胺诱导小鼠急性肝损伤模型;采用试剂盒测定血清转氨酶、脂质过氧化酶、抗氧化酶;采用H&E和免疫组织化学法检测组织病理学的改变;采用Western blot法检测凋亡通路的表达。结果啤酒花总黄酮各组的小鼠血清中ALT酶以及AST酶的数量都显著性下降;啤酒花总黄酮不仅可以降低肝组织中脂质过氧化物的含量,还能够升高超氧化物岐化酶(superoxide dismutase,T-SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)的活力;通过免疫组织化学法得出的结果是啤酒花总黄酮可以显著性地抑制NF-κB受体阳性反应产物在肝细胞中的表达;通过Western blot法得出的结果是啤酒花总黄酮可以抑制Bax蛋白的表达,提高Bcl-2蛋白的表达。结论啤酒花总黄酮对急性肝损伤具有一定的保护作用。
Objective To investigate the protective effect of hops flavonoids on thioacetamide-induced acute liver injury in mice. Methods The model of acute liver injury was induced by thioacetamide in mice. Serum aminotransferase, lipid peroxidase and antioxidant enzyme were detected by kit. H & E and immunohistochemistry were used to detect histopathological changes. Western blot Apoptotic pathway expression. Results The quantity of ALTase and AST enzyme in the serum of mice in each group of hops total flavonoids decreased significantly. The total flavonoids in hops not only reduced the contents of lipid peroxides in liver tissue, but also increased superoxide dismutase superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-PX). The results of immunohistochemistry showed that the total flavones of hop can significantly inhibit the activity of NF-κB The positive reaction product was expressed in hepatocytes. The result of Western blot showed that the total flavones of hop can inhibit the expression of Bax protein and increase the expression of Bcl-2 protein. Conclusion The total flavonoids of hops have some protective effects on acute liver injury.
