目的:研究人外周血中内皮祖细胞的生物学特性和诱导分化条件。方法:从健康成人外周血中分离内皮祖细胞接种于添加VEGF,bFGF,IGF-1,EGF的M199培养基中,观察细胞集落、梭形贴壁细胞的出现时间和特异性细胞标志表达情况。结果:第3~4天可观察到贴壁梭型细胞,第5~8天出现多个细胞团,第10天左右可见首尾相连形成条索状结构。贴壁细胞表达血管内皮细胞特异性标志。结论:成人外周血中的内皮祖细胞来源于单个核细胞,在一定条件下可以诱导分化为内皮细胞。
Objective: To study the biological characteristics and differentiation conditions of endothelial progenitor cells in human peripheral blood. METHODS: Endothelial progenitor cells were isolated from healthy adult peripheral blood and inoculated into M199 medium supplemented with VEGF, bFGF, IGF-1 and EGF. The appearance time of cell colonies and spindle-shaped adherent cells and the expression of specific cell markers were observed. Results: Adherent spindle cells were observed on the 3rd to 4th days. Multiple cell clusters appeared on the 5th to 8th day, and the cord-like structure was formed around the 10th day. Adherent cells express vascular endothelial cell specific markers. Conclusion: EPCs in adult peripheral blood are derived from mononuclear cells and can differentiate into endothelial cells under certain conditions.
