目的:研究槐耳清膏对人肺腺癌A2细胞的增殖抑制作用及凋亡促进作用,并探讨其分子机制。方法:MTT法检测槐耳清膏对细胞增殖的抑制作用;流式细胞仪检测对细胞凋亡的影响;Western blotting测定Wnt-1和Survivin蛋白表达的变化。结果:槐耳清膏能显著抑制人肺腺癌A2细胞增殖、促进凋亡,P<0.05;下调靶基因蛋白表达水平。结论:槐耳清膏可抑制人肺腺癌A2细胞增殖、促进细胞凋亡,其机制可能与下调Wnt-1和Survivin表达有关。
OBJECTIVE: To study the inhibitory effect and the promoting effect of Huaier clear paste on the proliferation of human lung adenocarcinoma A2 cells and to explore its molecular mechanism. Methods: MTT assay was used to detect the inhibitory effect of Huaqiao Ointment on cell proliferation. Flow cytometry was used to detect the effect on cell apoptosis. Western blotting was used to detect the protein expression of Wnt-1 and Survivin. Results: Huaier clear paste can significantly inhibit the proliferation of human lung adenocarcinoma A2 cells and promote apoptosis, P <0.05; down-regulate the expression of target gene protein. Conclusion: The Hua-hui cream can inhibit the proliferation of human lung adenocarcinoma A2 cells and promote cell apoptosis. The mechanism may be related to the down-regulation of Wnt-1 and Survivin expression.
