目的探究Smad4、转化生长因子-β1(TGF-β1)、转化生长因子-β受体1((TGF-βR1)在乳腺癌细胞中的表达和意义。方法选取2014年1月-2016年1月在该院确诊为乳腺浸润性导管癌并行手术治疗的女性患者60例,收集其标本作为观察组。另取正常乳腺组织20例作为对照组。采用免疫组化技术检测组织中的Smad4、TGF-β1和TGF-βR1,比较乳腺浸润性导管癌和正常乳腺组织中上述指标的表达差异及影响表达的因素。结果观察组TGF-β1和TGF-βR1的阳性表达率高于对照组,Smad4阳性表达率低于对照组,差异均有统计学意义(均P<0.05)。Smad4、TGF-β1和TGF-βR1在乳腺癌中的表达与年龄无关,而同分化程度和TNM分期有关,分化程度低、TNM分期高的标本TGF-β1和TGF-βR1的阳性表达程度高于分化程度高、TNM分期低的标本;分化程度低、TNM分期高的标本Smad4的阳性表达程度低于分化程度高、TNM分期低的标本,差异均有统计学意义(均P<0.05)。TGF-β1和TGF-βR1在乳腺癌组织中的表达呈正相关(r=0.647,P<0.05),而Smad4与TGF-β1、TGF-βR1的表达均呈负相关(r=-0.329和-0.426,均P<0.05)。结论 TGF-β1和TGF-βR1在乳腺浸润性导管癌组织中表达显著增加,而Smad4表达下降,表达程度均与分化程度和TNM分期有关。
Objective To investigate the expression and significance of Smad4, transforming growth factor-β1 (TGF-β1) and transforming growth factor-β receptor 1 (TGF-βR1) in breast cancer cells.METHODS From January 2014 to January 2016 Sixty female patients diagnosed as breast invasive ductal carcinoma with concurrent surgery were collected in this hospital and their specimens were collected as observation group.Other normal breast tissues were taken as control group.The immunohistochemistry was used to detect the expression of Smad4 and TGF- β1 and TGF-βR1 in invasive ductal carcinoma and normal breast tissue.Results The positive expression rates of TGF-β1 and TGF-βR1 in the observation group were higher than those in the control group, the positive expression of Smad4 (P <0.05) .Smad4, TGF-β1 and TGF-βR1 expression in breast cancer has nothing to do with the age, but with the degree of differentiation and TNM staging, the degree of differentiation is low The positive expression of TGF-β1 and TGF-βR1 in specimens with high TNM stage was higher than those with high differentiation and low TNM stage. The positive expression of Smad4 in specimens with low TNM stage and TNM stage was lower than that in TNM stage, Staging low specimens, the difference is (All P <0.05) .The expression of TGF-β1 and TGF-βR1 in breast cancer tissues was positively correlated (r = 0.647, P <0.05), while the expression of Smad4, TGF-β1 and TGF- (R = -0.329 and -0.426, all P <0.05) .Conclusion The expression of TGF-β1 and TGF-βR1 in breast invasive ductal carcinoma tissues increased significantly, while the expression of Smad4 decreased, the expression levels were correlated with the differentiation degree and TNM Staging related.
