目的:探讨青蒿琥酯(Art)抑制卵巢癌细胞株(CAOV3)增殖及p38通路的影响及机制。方法:培养卵巢癌细胞株(CAOV3),以四甲基偶氮唑盐(MTT)比色法及克隆形成实验测定Art抑制卵巢癌细胞株CAOV3增殖的作用。免疫印记法测定p-p38、Cyclin D1及p21waf/cip1表达。免疫沉淀法纯化蛋白并p38试剂盒测定p38活性。结果:MTT实验及克隆分析法均显示Art明显抑制CAOV3细胞增殖,Art抑制p-p38表达及活性、下调Cyclin D1表达,同时上调p21waf/cip1表达。结论:青蒿琥酯可以抑制卵巢癌细胞株(CAOV3)增殖,其机制与影响p38通路及下游Cyclin D1、p21waf/cip1表达有关。
Objective: To investigate the effect and mechanism of Artesunate on proliferation and p38 pathway in ovarian cancer cell line (CAOV3). Methods: The ovarian cancer cell line (CAOV3) was cultured. The effect of Art on the proliferation of ovarian cancer cell line CAOV3 was determined by MTT assay and colony formation assay. The expressions of p-p38, Cyclin D1 and p21waf / cip1 were detected by immunoblotting. The protein was purified by immunoprecipitation and the p38 activity was determined by the p38 kit. Results: MTT assay and clone analysis showed that Art significantly inhibited the proliferation of CAOV3 cells. Art inhibited the expression and activity of p-p38, down-regulated the expression of Cyclin D1 and up-regulated the expression of p21waf / cip1. Conclusion: Artesunate can inhibit the proliferation of ovarian cancer cell line (CAOV3), and its mechanism is related to the influence of p38 pathway and the expression of Cyclin D1 and p21waf / cip1.
