目的分析慢性丙型肝炎(CHC)患者外周血单核细胞亚群分布,同时观察负性调节分子T细胞免疫球蛋白及黏蛋白域蛋白3(Tim-3)和程序性细胞死亡蛋白1(PD-1)在各亚群表达的变化及意义。方法利用流式细胞术检测CHC患者外周血单核细胞亚群比例和各亚群Tim-3、PD-1表达,并与临床指标如丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)采用Spearman方法进行相关性分析。结果与健康对照组比较,CHC患者外周血CD14~+CD16~+单核细胞比例增高,以CD14CD16~+单核细胞比例增高显著。Tim-3在CD14CD16-单核细胞和CD14~+CD16单核细胞上表达水平升高;PD-1在CD14CD16-单核细胞和CD14CD16~+单核细胞上表达水平升高。CHC患者单核细胞亚群以及各亚群Tim-3、PD-1和临床指标ALT及AST无明显相关性。结论 Tim-3和PD-1在不同单核细胞亚群表达水平不同。
Objective To analyze the distribution of peripheral blood mononuclear cells in patients with chronic hepatitis C (CHC) and to observe the relationship between the expression of T-cell immunoglobulin and mucin domain protein 3 (Tim-3) and programmed cell death protein 1 -1) in all subgroups and its significance. Methods Flow Cytometry was used to detect the expression of Tim-3 and PD-1 in peripheral blood mononuclear cells (PBMCs) in CHC patients. The expressions of Tim-3, Transferase (AST) using Spearman method for correlation analysis. Results Compared with healthy control group, the proportion of CD14 ~ + CD16 ~ + monocytes in CHC patients increased, and the proportion of CD14 + CD16 ~ + monocytes increased significantly. Expression of Tim-3 on CD14CD16- monocytes and CD14 ~ + CD16 monocytes increased; PD-1 was expressed on CD14CD16- monocytes and CD14CD16 + monocytes high. CHC patients with monocyte subsets, as well as Tim-3, PD-1 and clinical indicators ALT and AST no significant correlation between each subpopulation. Conclusion The expression of Tim-3 and PD-1 in different monocyte subsets is different.
