目的:观察低剂量辐射外周血单个核细胞(PBMC)联合左旋棉酚对体外培养人前列腺癌LNCaP细胞的杀伤作用。方法:采用l Gyγ射线照射人PBMC,辐射剂量率为17Gy/min,实验设对照组、照射及未照射的PBMC与LNCaP细胞共培养组、左旋棉酚处理组及照射的PBMC与左旋棉酚联合作用组。利用吖啶橙/溴化乙啶(AO/EB)荧光双染色法及MTT法,观察PBMC和/或左旋棉酚对肿瘤细胞的杀伤情况。结果:辐照的PBMC及左旋棉酚处理组对LNCaP细胞的杀伤作用明显增强,杀伤活性明显高于未辐照组(P<0.05),而联合作用组的杀伤活性明显高于辐照组及左旋棉酚处理组(P<0.01)。结论:低剂量辐射PBMC联合左旋棉酚可以明显提高其体外的抗肿瘤效应。
Objective: To observe the killing effect of low dose radiation of peripheral blood mononuclear cells (PBMCs) combined with L - gossypol on human prostate cancer LNCaP cells. Methods: Human peripheral blood mononuclear cells (PBMCs) were irradiated with 1 Gy γ-rays at a dose rate of 17 Gy / min. In the experimental group, irradiated and non-irradiated PBMCs were co-cultured with LNCaP cells and treated with L-galactose. Role group. Conclusion: Low dose radiation PBMC combined with L-Gossypol can significantly enhance its anti-tumor effect in vitro.
