目的研究海军潜水人群铜绿假单胞菌分离株的遗传基因型,明确海军潜水人群铜绿假单胞菌携带株的流行特征。方法用多位点序列分型(multilocus sequence typing,MLST)技术、以铜绿假单胞菌7个管家基因acsA、aroE、guaA、mutL、nuoD、ppsA和trpE内部特定核酸片段作检测靶,对随机抽样海军潜水员携带的64株铜绿假单胞菌进行目的基因扩增和测序,应用Pseudomonas aeruginosa MLST数据库对测序结果进行分析,以获得检测靶基因的多位点序列信息;应用Bionumerics 4.0的LIAN,SplitsTree和eBURST分析程序对获得的MLST进行进一步的生物信息学分析,以获得流行株的遗传特征。结果 64株铜绿假单胞菌中53株可分为19个ST型,11株未能分型;其中ST274和ST260是优势克隆株,分别占18.75%(12/64)、15.62%(10/64)。结论海军潜水人群携带的铜绿假单胞菌具有遗传多样性和优势基因型,MLST分型对于研究海军潜水人群携带的铜绿假单胞菌遗传差异与流行特征具有重要意义。
Objective To study the genotypes of Pseudomonas aeruginosa isolates in the submarine of the Navy and determine the prevalence of Pseudomonas aeruginosa carriers in the Navy. Methods Using specific multilocus sequence typing (MLST) technique, specific fragments of acsA, aroE, guaA, mutL, nuoD, ppsA and trpE in 7 housekeeping genes of Pseudomonas aeruginosa A total of 64 Pseudomonas aeruginosa samples taken from the sampled naval divers were amplified and sequenced. The Pseudomonas aeruginosa MLST database was used to analyze the sequencing results to obtain multi-locus sequence information of the target gene. The Bionumerics 4.0 LIAN, SplitsTree And eBURST analysis program for further bioinformatics analysis of the obtained MLST to obtain the genetic characteristics of the epidemic strains. Results 53 strains of 64 Pseudomonas aeruginosa strains could be divided into 19 ST types and 11 strains could not be typed. ST274 and ST260 were dominant clones, accounting for 18.75% (12/64) and 15.62% (10 / 64). Conclusion The Pseudomonas aeruginosa carried by submarine naval populations has the genetic diversity and predominant genotype. MLST typing is of great significance for studying the genetic diversity and epidemiological characteristics of Pseudomonas aeruginosa carried by navy diving population.
